From the Main Menu, use you Left or Right button to highlight the pot below the menu card. Press your Enter button. The green vial will pop up and be caught by Yzma, who turns Flo into Kuzco.

Interface:
From the Main Menu, use your Right or Left button to highlight to the pot on the far left of the screen. Press your Enter button. The pot will explode drenching the screen in cheese goo.
